چکیده

In the earlier papers in this series (3, 4) data have been presented which indicate tha t the isolated perfused liver from a ra t fed 8'-methyldimethylaminoazobenzene 1 has a decreased ability to oxidatively catabolize amino acids. Such livers, however, show an increased capacity for incorporation of C14-1abeled amino acid into both liver and plasma proteins. A biochemical block or impairment in the chemical reaction sequence from amino acid nitrogen to urea nitrogen was thought to develop during the process of azo dye carcinogenesis. To a t t empt to demonstrate definitively the locus of this metabolic impairment, we have studied the ability of the perfused livers from 3'-Me-DABfed rats to produce urea from ammonia, aspartic acid, or arginine added to the blood perfusate.

Effect of azo-dye carcinogenesis on enzymes concerned with urea synthesis in rat liver.

Dickens & Weil-Malherbe (1943) first demonstrated that slices of liver tumours, induced by giving DAB* in the diet, when incubated in vitro with ammonium salts and alanine in the presence of trace amounts of omithine, were unable to synthesize urea and that in this respect the tumour had lost one of the specialized functions of normal liver. The work of Greenstein (1942), who used transplanted ...

The biosynthesis of guanidinosuccinic acid by perfused rat liver.

The metabolic pathway for the synthesis of guanidinosuccinic acid was studied in the rat. Labeled guanidinosuccinic acid was isolated from the urine of animals given L-[guanidino-14C]arginine intraperitoneally but did not appear in the urine after administration of D,L-[guanidino-14C]canavanine. Radioactive arginine and nonradioactive aspartic acid and arginine were infused in the isolated, per...

Effects of Confinement/crowding on Ureogenesis in the Gulf Toadfish Opsanus Beta

In order to elucidate the cues for, and the biochemical mechanisms of, the transition to ureogenesis in the gulf toadfish Opsanus beta, experiments on the effects of confinement/crowding were carried out. Confinement of toadfish to small volumes of water initiated a switch to nearly complete reliance on ureogenesis for nitrogen excretion within 2448 h. Further experiments suggested that th...

The Hepatoprotective Effects of Corn Silk against Dose-induced Injury of Ecstasy (MDMA) Using Isolated Rat Liver Perfusion System

Background: Corn silk (CS) is widely used in Iranian traditional medicine. The aim of this study was to investigate hepatoprotective activity of CS by Isolated Rat Liver Perfusion System (IRLP). Methods: Hydro-alcoholic extract of corn silk (10, 20, 40, and 100 mg kg-1) was evaluated for its hepatoprotective activity by IRLP. Phenol and flavonoid contents of the extract were determined as gal...

Role of the glutamate dehydrogenase reaction in furnishing aspartate nitrogen for urea synthesis: studies in perfused rat liver with 15N.

The present study was designed to determine: (i) the role of the reductive amination of alpha-ketoglutarate via the glutamate dehydrogenase reaction in furnishing mitochondrial glutamate and its transamination into aspartate; (ii) the relative incorporation of perfusate 15NH4Cl, [2-15N]glutamine or [5-15N]glutamine into carbamoyl phosphate and aspartate-N and, thereby, [15N]urea isotopomers; an...
